The University of Siena (UNISI)

The University of Siena (UNISI) was founded in 1240, thus being one of the ancient italian University. Today UNISI is still among the most important Italian campus, mostly due to the fame of its faculties of law and medicine, according to recent results of national and international rating agencies.

The Department of Molecular Biology of UNISI is a scientific structure that promotes and coordinates research and teaching activities in the fields of Biochemistry, Biophysics, Molecular and Cellular Biology, Biotechnology, Pharmacology, Genetics, Infectivology and Microbiology. It is formed by several laboratories equipped with both basic and advanced facilities to perform molecular analysis work. Principal fields of investigation of the Microbiology Section are: 1) antimicrobial resistance surveillance; 2) molecular epidemiology and biochemical characterization of resistances; 3) characterization of genetic supports and mobility of resistance determinants; 4) advanced molecular methods for detection of resistance determinants. Personnel of Microbiology Section has a notable experience and knowledge in the investigation of resistance mechanisms of pathogenic bacteria and therefore can give a precious contribute in the development of TEMPOTest-QC project. The Department houses the most common microbiology and molecular biology instruments, with the following specific platforms:

• Microarray platform (arrayer and scanner) (Affymetrix).
• 2 UV-VIS Cary 100 spectrophotometers (Varian Inc.).
• Electroporation device (Bio-Rad).
• FPLC chromatography protein purification platform (Pharmacia Biotech).
• AKTA Purifier chromatography protein purification platform (Amersham Biosciences).
• Mini protean III electrophoresis system (Bio-Rad).
• LKB bidimensional electrophoresis system + cryostat (LKB-Pharmacia).
• Labsonic 1000L Sonicator (B. Braun).
• Digital refrigerated incubator shaker (New Brunswick Scientific Inc.).
• Thermal cycler Light Cycler (Roche) for capillary-based RT-PCR.
• Thermal cycler DNA Engine Opticon (MJ Research) for microplate-based RT-PCR.
• Thermal cyclers (MWG Biotech, Bio-Rad, Perkin Elmer).
• Perkin Elmer Janus Robotic liquid-handling System, integrated with a multi-technology microplate reader (UV-Vis spectrophotometry, fluorescence, time-resolved fluorescence, polarized fluorescence, luminescence).
